IV. Specific accomplishments in research

Prof. Alonso pioneered the design and development of novel nanostructures based on biopolymers intended to the targeted delivery of drugs, notably biological drugs. More specifically:

PLGA and PLA-PEG nanoparticles for the encapsulation and controlled release of proteins, antigens and polynucleotides from biodegradable were first reported in 1997/98. This achievement has had a great impact in the peptide/protein delivery field and currently a high number of biotech and pharma companies are developing products based on these technologies. It was also proven for the first time the influence of the polymer’s PEGylation on the capacity of nanoparticles (PLA-PEG) to overcome mucosal barriers (ocular, nasal, intestinal) and to facilitate the transport of peptides, proteins and polynucleotides across them. This discovery has been followed by substantial literature in the area o muco-penetration of nanoparticles.

In the field of vaccination starting in the 90’s at MIT, she has collaborated with the World Health Organization, the Bill & Melinda Gates Foundation, the NIH and the European Comission in the development of needle-free vaccination strategies for a number of vaccines. Her hast contribution has been on an HIV vaccine candidate, currently in advanced preclinical development. Currently, she is working in a series of mRNA nasal vaccines in the context of a large European project NOSEVAC (https://www.euvaccine.eu/post/nosevac-a-new-european-collaboration-to-develop-nasal-vaccines).

Polysaccharide chitosan and its combinations with hyaluronan or dextran, for the delivery of drugs and vaccines. The first reports on these delivery carriers were published in 1997. The scientific impact of this work can be measured in terms of thousands of articles on chitosan nanoparticles. These technologies have been adapted for the nasal and intestinal absorption of peptides, for nasal vaccination and for the delivery of RNA to the eye. This knowledge has led to the start-up company “Smart Vitamins”. In addition, in collaboration with the company Privo technologies, this technology has led a new treatment for head and neck cancer (Phase 3 clinical trials) and other types of cancer.

Multifunctional Polymeric Nanocapsules (MPN) technology first reported in 2,000 and adapted to the intracellular delivery of monoclonal antibodies intended to reach oncoprotein target that are so far undrugged. This discovery has led to the starting of Libera Bio, a spin-off company from the University of Santiago de Compostela (USC).

Overall, Prof. Alonso’s discoveries led to significant clinical advances in the development of new treatments and vaccinations strategies.
